Effect of specific growth rate on the production of a recombinant nuclease by Escherichia coli

چکیده

The effect of specific growth rate μ on the production of Vibrio vulnificus nuclease was investigated in fed-batch cultures of a high-yield recombinant Escherichia coli. A pseudo-exponential feeding method for controlling μ was employed to obtain the desired data. It was found that the nuclease content of cells γP varied with μ, with a maximum occurred at μ = 0.05 h−1; however, the specific nuclease production rate qp increased with increasing μ. The synthesis of nuclease was dependent on μ, irrespective of the feeding methods. When concerning the compromise between the nuclease yield and its production rate, the linear-gradient feeding method, being simple and adaptable, was shown to be adequate for the nuclease production. © 2003 Elsevier Science B.V.
